fast pyrolysis of biomass to produce bio-oil followed by catalytic enhancement is widely studied approach to the potential for fuel production from biomass. Because of the complexity of the bio-oil, most of the strategy to increase focus on eliminating oxygen from the whole mixture for fuel production.
Here we report a new method for the production of specialty chemicals, acids gluconate, from pyrolysis of biomass. Through a combination of sequential condensation of steam pyrolysis and extraction of water, levoglucosan rich solution is obtained that accounts for over 30% of the carbon in the bio-oil produced from red oak. A simple filtration step produces high-purity stream levoglucosan. This flow levoglucosan then hydrolyzed and partially oxidized to produce gluconic acid with high purity and selectivity.
The combination of cost-effective pyrolysis coupled with simple separation and upgrades can enable a variety of new product market for chemicals from biomass. In this review, we discuss recent advances in the field of synthetic biology, and describe how these tools have been applied to produce a wide variety of chemicals in microorganisms. Here we classify the expansion of synthetic biology toolbox into three different categories based on their main functions in the tension-engineering for the design, construction, and for optimization.
Next, focus on the last few years, we see how the chemicals have been produced using the tools of the new synthetic biology. Progress in producing fuel is briefly described, followed by a more thorough treatment of commodity chemicals, specialty chemicals, pharmaceuticals, and nutraceuticals. Throughout this review, emphasis is placed on how the tools of synthetic biology applied to strain engineering. Finally, we discuss the host organism and strain diversity and provides future prospects in the field.

Engineering a biological conversion of methanol for specialty chemicals in Escherichia coli.
Methanol is an attractive substrate for the biological production of chemicals and fuels. Mechanical methylotrophic organism Escherichia coli as a platform for converting methanol to the desired metabolite. Prior efforts to engineer E. coli methylotrophic bounded by methanol dehydrogenases (MDHS) with enzyme kinetics that are not profitable.
We engineered E. coli to utilize methanol using superior NAD dependent MDH of Bacillus stearothermophilus and ribulose monophosphate (butt) enzyme pathways of B. methanolicus. Using 13C-labeled, we show this E. coli strain converts methanol into biomass components. For example, the key intermediates of the TCA cycle, succinate and malate, exhibit labels up to 39%, while the lower glycolytic intermediate, 3-phosphoglycerate, up to 53%. Some carbon labeled for each compound, showing the butt cycling track for methanol assimilation to support growth.
